تهیه جهش یافته های مقاوم به آنتی بیوتیک باکتریPseudomonas fluorescens و بررسی فعالیت و مکانیسمهای آنتاگونیستی آنها بر علیه قارچ Rhizoctonia solani عامل بیماری مرگ گیاهچه پنبه

چکیده

در این تحقیق با استفاده از پدیده جهش خود به خودی(Spontaneous mutation) سه جهش یافته مقاوم به دو آنتی بیوتیک (Nalidixic acid (Nal  و Rifampicin (Rif)  از شش جدایه باکتری Pseudomonas fluorescens ،تهیه گردید و در مقایسه با انواع وحشی آنها در شرایط آزمایشگاه بر علیه قارچ بیماریزایRhizoctonia solaniعامل بیماری مرگ گیاهچه پنبه از طریق روش آزمون کشت متقابل (Dual culture)مورد بررسی قرار گرفت. نتایج نشان داد که دو جدایه از سه جدایه جهش یافته خصوصیات آنتاگونیستی بهتری نسبت به انواع وحشی خود نشان دادند . همچنین جدایه های جهش یافته از نظر مکانیسمهای بازدارندگی بر علیه قارچ مذکور مورد بررسی قرار گرفته و نتایج حاکی از آن بود که هر سه باکتری جهش یافته ازنظر تولید مواد فرار بهتر از انواع وحشی ، از نظر تولید مواد غیرفرار تقریبا مشابه و از نظر تولید سیدروفور برخی جدایه های جهش یافته در برخی غلظت ها نسبت به انواع وحشی آنها توانستند سیدروفور بیشتری تولید کنند . نتایج این تحقیق نشان دهنده این است که پدیده جهش خودجوش ممکن است بر خواص آنتاگونیستی باکتریهای آنتاگونیست تاثیر گذاشته و آن را افزایش دهد.

In this study, antibiotic resistant mutants of antagonistic bacteria to Rifampicin and Nalidixic acid using spontaneous mutation were obtained. Antagonistic activity of three isolates belonging to Pseudomonas fluorescens were tested in vitro against a virulent isolate of Rhizoctonia solani, the causal agent of cotton seedling damping-off, using dual culture method. It was shown that two isolates had better antagonistic traits against fungus in comparison with their wild types. The above isolates were also evaluated for their inhibitory mechanisms against fungal pathogen and the results showed that all three mutants were different than their wild types in volatile metabolites production. There were no differences between the mutants and wild types in production of non-volatile metabolites. However, some mutants (Pf-C-1, Pf-W-2) produced more siderophores than their wild types. Results of this study suggest that spontaneous mutation may affect and enhance the activity and performance of antagonistic bacteria.
